Which is the best bike in the 350-400cc segment if I'm looking for mileage, comfort, low maintenance, and long-term ownership? I ride around 500km every week.

For your use case, we would recommend looking at the Honda CB350 models, Royal Enfield Bullet/Classic 350, and Triumph Speed T4. All of these 350cc machines set a high bar for comfort, but the Bullet and Classic will definitely have a small edge here. When it comes to fuel economy, the CB350s and Speed T4 will likely be more efficient than the heavier Royal Enfield bikes, although mileage varies greatly depending on a multitude of factors. Finally, when it comes to long-term peaceful ownership, it's likely that the Hondas will be the easiest to live with, although maintenance costs for all three will be pretty comparable.
These three options are also very different in terms of their riding experience, and to accurately gauge which one will be best for you, we'd recommend a test ride of all before you make your decision.

@hassanahmed Go for CB350 because Honda engines are a class apart. Very smooth gearbox as well. And you don't feel the weight of the bike at all.

@hassanahmed Go for Classic 350 because it's the classic . The cult plus the service network and spares availability . A product is only as good as this.

Does maintenance of t4 speed is high then cb350 or same ? Kindly suggest.. im planning for t4 but im worried about mileage and maintenance service cost. As i heard cb350 is low.

@chin The Triumph Speed T4 isn't significantly more expensive to maintain than the Honda CB350, though the CB350 may have a slight edge in service costs and fuel efficiency. I wouldn't let maintenance be the deciding factor - if you prefer the Speed T4's performance and riding experience, it's a worthwhile choice.
